High-Resolution DTI of Human Articular Cartilage with Long Diffusion Time: Preliminary Findings

چکیده
NTRODUCTION The articular cartilage has a matrix structure of three layers superficial zone, middle zone and deep zone, which are divided by the different orientations of collagens. Recent publications have shown fractional anisotropy (FA) can reflect the orientation of the collagen fibers using diffusion tensor imaging (DTI) in MRI [1-3]. However, the FA contrasts were low especially between the superficial zone and middle zone in those literatures. This was possibly because they selected the short diffusion time (Δ < 10 ms) [1-3]. In our study, we chose both Δ = 10 ms and Δ = 30 ms and kept the same b = 500 s/mm in six directions of diffusion gradients. The FA contrast between superficial zone and middle zone in Δ = 30 ms increased by about 4 times of the FA contrast in Δ = 10 ms. After increasing Δ, the mean diffusivity (MD) in each zone decreased because MD depended not only on the self diffusion of fluid in cartilage but also on the pore size [4], while the maximum diffusivity (MaxD) kept almost unchanged since MaxD was along the principal diffusion direction and experienced less restricted diffusion. Our preliminary data shows the MaxD has the potential to be used as a marker of the proteoglycans (PGs) concentration. Future development of this approach may be important for detecting the early degeneration of cartilage.

Background: Previous studies have shown that Galectin-3, a member of lectin family, is expressed in developing cartilage in mouse embryo and also in growth plate of long bones. Objective: In the present work, the expression pattern of Galectin-3 in normal and various grades of osteoarthritic (OA) human articular cartilage has been studied. Methods: Using immunohistochemistry and standard we...

Materials and Methods: Articular cartilage was harvested from knee joints of 10 male rats and was digested using enzymatic solution consisting of 0.2% collagenase I and 0.1% pronase. Released chondrocyte were then plate...
متن کاملDetermination of Fiber Direction in High Angular Resolution Diffusion Images using Spherical Harmonics Functions and Wiener Filter
Diffusion tensor imaging (DTI) MRI is a noninvasive imaging method of the cerebral tissues whose fibers directions are not evaluated correctly in the regions of the crossing fibers. For the same reason the high angular resolution diffusion images (HARDI) are used for estimation of the fiber direction in each voxel. One of the main methods to specify the direction of fibers is usage of the spher...
متن کاملCD147 (Extracellular Matrix Metalloproteinase Inducer-EMMPRIN) Expression by Human Articular Chondrocytes
Background: Integrins are a family of transmembrane proteins that allow communication between the extracellular matrix and the interior of cells. Chondrocytes, cells of articular cartilage, express integrins and these molecules appear to have a variety of roles including mechanotransduction. Integrins are known to associate with a number of accessory molecules such as CD147 that may act to regu...
